Résumé / Abstract

A simple procedure is presented based on simultaneous plasma deproteinization and derivatization with acetic anhydride for the determination of metformin in human plasma by liquid chromatography. The analyte and the internal standard (methyl biguanidine) remained unchanged after the derivatization reaction, and are soluble in the final supernatant. An ion-pair mechanism was applied for their separation and ultraviolet detection was carried out at 232 nm. Elution was achieved with a mobile phase consisting in 21% acetonitrile and 79% aqueous component (containing 0.01 moles/L sodium octanesulphonate brought to pH = 2.5 with H3PO4), at a flow-rate of 1 mL/min. A double end capped, 5 microns particle size Inertsil ODS-2 stationary phase (150 mm length, 4.6 mm internal diameter column) was used. Detection limit of 32.5 ppb was obtained. The procedure was applied to a bioequivalence study (22 healthy volunteers) of two drugs containing 850 mg metformin.
