Evaluation of gentamicin-Eudragit microspheres as ophthalmic delivery systems in inflamed rabbit's eyes

Gentamicin sulfate microspheres were prepared from poly methyl methacrylate (Eudragit RS and RL) polymers, cross-linked with polyvinyl alcohol, using a modified double emulsion-solvent evaporation technique. Successful trials were done to investigate the effectiveness of the prepared gentamicin-Eudragit microspheres in the treatment of inflamed rabbits' eyes. The healing effect of gentamicin-microspheres was compared with the marketed gentamicin eye drops and drug free particles. The healing rates of gentamicin formulations were treated as first-order kinetics from which the healing rate constants (k) and healing half-lives (t
1/2) were calculated. The results revealed that gentamicin-Eudragit microspheres have accelerated the rate of healing and reduced the healing half-life in comparison with the marketed gentamicin eye drops. Drug free particles gave comparable results with the control studies. The calculated healing half-lives for the gentamicin formulations were found to be 9 and 27 days for the microspheres and eye solution, respectively. The results of the statistical analysis assure the increase in therapeutic effectiveness and corneal retention of the developed ophthalmic drug delivery system.
